• Series of ionenes with substantial bioactivity against model microorganisms.
• An unusual drop of activity following increase of polymer hydrophobicity.
• Vesicles containing cardiolipin are sensitive toward more hydrophilic ionenes.
• For certain vesicles and concentrations hydrophobic ionenes can reduce release of a dye.
